Research:

Our lab studies the hormone-mediated transcription of genes controlling steroidogenesis in the ovary. One of our main interests had been in the Steroidogenic acute regulatory (StAR) protein gene. StAR mediates the rate-limiting step of cholesterol delivery to the mitochondrion for steroid synthesis. Our previous work cloned the LDL receptor and StAR gene promoters in the pig and showed that both were responsive to the hormones FSH and LH in granulosa cells. The gonadotropin effects on these promoters were augmented by IGF-I. Further analysis of the StAR promoter identified GATA4 and C/EBPbeta as co-mediators of the FSH/IGF-I effect. We found that FSH stimulated the phosphorylation of GATA4 on a PKA site. These previous studies have led to our current work with GATA4. Our goal is to identify the true GATA4 responsive gene pool(s) during follicular and luteal development. Along these lines we are studying the role of phosphorylation in GATA4 transactivation.

StAR expression is suppressed in the granulosa layer during follicular development. One of interests is to determine how StAR remains quiescent during this period. These studies have led us to investigate histone modifications within the StAR gene that may keep it silent before the midcycle gonadotropin surge that causes ovulation and terminal differentiation of the follicular cells into luteal cells.
